    Hello all,

Wondering what temperature others are incubating their anaerobic plates at... 21°C? 28°C? 35°C?

    Thank you,

    Adam Donkers

    Hi Adam,

We usually target between 32C and 37C, depending on the type of media and organisms you are looking for.

I've always used 28°C for aerobic and anaerobic plates. I think we had a small incubator at 35°C that was only used for testing process water samples.

General recommendation for incubation temperature according to EBC methods is 25 degrees plus/minus 2.

This temperature is good for lactic acid bacteria as well as for the strict anaerobes and wild yeast growth.

Only for diastaticus specific testing, higher temperatures of 30 degrees up to 37 degrees are recommended.

Some lactics and also yeasts won't grow at 30 degrees and higher.
